Alteration of the Reproductive Patterns in Drosophila melanogaster by Effects of High Concentrations of Boron on In Vitro Cultured Medium International Journal of Morphology
Espinoza-Navarro,O; Rodríguez,H; Rodríguez,M; Silva,E; Luque,A.
The objective of this study was to evaluate the effects of boron (B) using as a biological model Drosophila malanogaster. Analizing specifically the responses over descendency to doses of 6,0 and 12 mg/L, comparing a control group which culture medium was prepared with water with boron concentrations of 0,6 mg/L, according to World Health Organization. The results show a significative alteration in the number of descendency from F1 to F3 and then a tendency to normalization in F4 and F5. Also a significant change in proportions between the males and females, with an acute effect on F1 and then a possible chronic effect from F2 to F5, increasing the proportion of females over males. This differences between genders are significant only until F4, on F5 these...

Are larger and/or more symmetrical Drosophila melanogaster (Diptera, Drosophilidae) males more successful in matings in nature? Rev. Bras. entomol.
Pavković-Lučić,Sofija; Kekić,Vladimir.
Are larger and/or more symmetrical Drosophila melanogaster (Diptera, Drosophilidae) males more successful in matings in nature? Sexual selection in Drosophila melanogaster, related to body size and fluctuating asymmetry in wing length and number of sex comb teeth in males, was tested in natural conditions. Males collected in copula were significantly larger than those collected as a single, while no difference in mean number of sex comb teeth between copulating and single males was observed. On the other hand, single males had greater asymmetry both for wing length and number of sex comb teeth than their mating counterparts. It looks like that symmetry of these bilateral traits also may play a role in sexual selection in this dipteran species in nature.

Distribution of the Bari-I transposable element in stable hybrid strains between Drosophila melanogaster and Drosophila simulans and in Brazilian populations of these species Genet. Mol. Biol.
Lucca Jr.,Marcos de; Carareto,Claudia Marcia Aparecida; Ceron,Carlos Roberto.
We analyzed the distribution of the Bari-I transposable element in Drosophila melanogaster (IN(1)AB), its sibling species Drosophila simulans (C167.4) and in eight hybrid strains derived from initial crosses involving D. simulans females and D. melanogaster males of the above cited strains as well as in Brazilian populations of these species. Polymerase chain reaction (PCR) data showed the presence of the Bari-I element among species populations and hybrid strains. Hybridization with a 703 bp probe homologous to the Bari-I sequence showed that the number of Bari-I copies in D. melanogaster IN(1)AB was higher than in D. simulans C167.4 strains. Hybrid strains presented Bari-I sequences related to both parental species. In addition some strains displayed a...

DNA damage protective effect of honey-sweetened cashew apple nectar in Drosophila melanogaster Genet. Mol. Biol.
Silva,Robson Alves da; Dihl,Rafael Rodrigues; Dias,Lucas Pinheiro; Costa,Maiane Papke; Abreu,Bianca Regina Ribas de; Cunha,Kênya Silva; Lehmann,Mauricio.
Abstract Fruits and derivatives, such as juices, are complex mixtures of chemicals, some of which may have mutagenic and/or carcinogenic potential, while others may have antimutagenic and/or anticancer activities. The modulating effects of honey-sweetened cashew apple nectar (HSCAN), on somatic mutation and recombination induced by ethyl methanesulfonate (EMS) and mitomycin C (MMC) were evaluated with the wing spot test in Drosophila melanogaster using co- and post-treatment protocols. Additionally, the antimutagenic activity of two HSCAN components, cashew apple pulp and honey, in MMC-induced DNA damage was also investigated. HSCAN reduced the mutagenic activity of both EMS and MMC in the co-treatment protocol, but had a co-mutagenic effect when...

Effect of organic tomato ( Lycopersicon esculentum ) extract on the genotoxicity of doxorubicin in the Drosophila wing spot test Genet. Mol. Biol.
Dutra,Elaine S.; Dias,Cristina D.; Araújo,Bethânia C. de; Castro,Antônio J. S.; Nepomuceno,Júlio C..
The consumption of organic tomatoes (ORTs) reduces the risk of harmful effects to humans and the environment caused by exposure to toxic agrochemicals. In this study, we used the somatic mutation and recombination test (SMART) of wing spots in Drosophila melanogaster to evaluate the genotoxicity of ORT and the effect of cotreatment with ORT on the genotoxicity of Doxorubicin ® (DXR, a cancer chemotherapeutic agent) that is mediated by free radical formation. Standard (ST) cross larvae were treated chronically with solutions containing 25%, 50% or 100% of an aqueous extract of ORT, in the absence and presence of DXR (0.125 mg/mL), and the number of mutant spots on the wings of emergent flies was counted. ORT alone was not genotoxic but enhanced the...

Effects of artichoke (Cynara scolymus) leaf and bloom head extracts on chemically induced DNA lesions in Drosophila melanogaster Genet. Mol. Biol.
Jacociunas,Laura Vicedo; Dihl,Rafael Rodrigues; Lehmann,Mauricio; Ferraz,Alexandre de Barros Falcão; Richter,Marc François; Silva,Juliana da; Andrade,Heloísa Helena Rodrigues de.
The genotoxicity of bloom head (BHE) and leaf (LE) extracts from artichoke (Cynara scolymus L.), and their ability to modulate the mutagenicity and recombinogenicity of two alkylating agents (ethyl methanesulfonate - EMS and mitomycin C - MMC) and the intercalating agent bleomycin (BLM), were examined using the somatic mutation and recombination test (SMART) in Drosophila melanogaster. Neither the mutagenicity nor the recombinogenicity of BLM or MMC was modified by co- or post-treatment with BHE or LE. In contrast, co-treatment with BHE significantly enhanced the EMS-induced genotoxicity involving mutagenic and/or recombinant events. Co-treatment with LE did not alter the genotoxicity of EMS whereas post-treatment with the highest dose of LE significantly...

Excitatory effects of Buthus C56 toxin on Drosophila larval neuromuscular junction J. Venom. Anim. Toxins incl. Trop. Dis.
Gawade,S. P..
Buthus C56 toxin from venom of the Indian red scorpion Mesobuthus tamulus was studied for its effects on spontaneous miniature excitatory junctional potentials (MEJP) on Drosophila larval neuromuscular junctions. C56 toxin was isolated on CM-Cellulose with linear gradient of ammonium acetate buffer, pH 6.0. Toxin purity was determined on SDS slab gel electrophoresis. Effective concentration of C56 toxin was based on contraction paralysis units (CPU) in Drosophila 3rd instar larvae by microinjection (0.1 CPU/ml = 2 x 10-6 g/ml). The toxin-induced excitatory junctional potentials were studied for calcium dependency (0.2 mM to 1.2 mM Ca2+) in Drosophila Ringer. Excitatory junctional potential amplitude was increased with increasing calcium concentration;...

Genotoxicity of lapachol evaluated by wing spot test of Drosophila melanogaster Genet. Mol. Biol.
Costa,Wender Ferreira; Oliveira,Alaide Braga de; Nepomuceno,Júlio César.
This study investigated the genotoxicity of Lapachol (LAP) evaluated by wing spot test of Drosophila melanogaster in the descendants from standard (ST) and high bioactivation (HB) crosses. This assay detects the loss of heterozygosity of marker genes expressed phenotypically on the fly's wings. Drosophila has extensive genetic homology to mammals, which makes it a suitable model organism for genotoxic investigations. Three-day-old larvae from ST crosses (females flr³/TM3, Bd s x males mwh/mwh), with basal levels of the cytochrome P450 and larvae of high metabolic bioactivity capacity (HB cross) (females ORR; flr³/TM3, Bd s x males mwh/mwh), were used. The results showed that LAP is a promutagen, exhibiting genotoxic activity in larvae from the HB cross. In...

Modulatory effects of the antioxidant ascorbic acid on the direct genotoxicity of doxorubicin in somatic cells of Drosophila melanogaster Genet. Mol. Biol.
Fragiorge,Edson José; Spanó,Mário Antônio; Antunes,Lusânia Maria Greggi.
In this study two different crosses involving the wing cell markers mwh and flr³ (standard (ST) cross and high bioactivation (HB) cross, the latter being characterized by a high constitutive level of cytochrome P450 which leads to an increased sensitivity to a number of promutagens and procarcinogens) were used to investigate the modulatory effects of ascorbic acid (AA) combined with the antitumor agent doxorubicin (DXR) in Drosophila melanogaster. We observed that the two different concentrations of AA (50 or 100 mM) had no effect on spots frequencies, while DXR treatments (0.2 or 0.4 mM) gave positive results for all types of spots, when compared to negative control. For marker-heterozygous (MH) flies, a protective effect was observed with the lower...

Phenotypic plasticity for Drosophila melanogaster (Diptera: Drosophilidae) larval olfactory behaviour in response to whole fruit olfactory stimuli Rev. Soc. Entomol. Argent.
Lavagnino,Nicolás J.; Fanara,Juan J..
Drosophila melanogaster Meigen 1830 (Diptera: Drosophilidae) is the quintessential insect model organism. However, with a few exceptions, ecological features of this species have been poorly investigated. In the present work we describe a behavioural assay to quantify olfactory behaviour of D. melanogaster larvae in response to complex olfactory stimuli that are present in the natural environments, i. e.: rotten fruits that act as hosts in nature. Results obtained using this assay reveal that there is intra-population genetic variation and phenotypic plasticity for the character in a natural population from west-central Argentina.

Protective effects of Berberís crataegina DC. (Ranunculales: Berberidaceae) extract on Bleomycin-induced toxicity in fruit flies (Diptera: Drosophilidae) Rev. Soc. Entomol. Argent.
ATICI,Tugba; ALTUN QOLAK,Deniz; ERSOZ,Qagla.
Antineoplastic agents due to their cytotoxic effects increase the amount of free radical in cells causing cell death. The aims of this study were to assess the possible toxic effects of Bleomycin (BLM) on the number of offspring and survival rate of fruit flies as well as the protective role of Berberis crataegina DC. fruit extracts (BCE) on these effects. Solutions prepared in different concentrations (0.125 mg/mL for BLM; 10 and 20 mg/mL for BCE) were applied to the adult fruit fly Drosophila melanogaster's wild-type individuals and 72 ± 4 h larvae to determine the number of offspring and survival rate, respectively. As a result, the higher number of offspring and survival rate in the BCE + BLM applied groups than the only BLM applied groups shows that...

Recombinogenic activity of Pantoprazole® in somatic cells of Drosophila melanogaster Genet. Mol. Biol.
Lopes,Jeyson Césary; Machado,Nayane Moreira; Saturnino,Rosiane Soares; Nepomuceno,Júlio César.
Pantoprazole® is one of the leading proton pump inhibitors (PPIs) used in the treatment of a variety of diseases related to the upper gastrointestinal tract. However, studies have shown an increased risk of developing gastric cancer, intestinal metaplasia and hyperplasia of endocrine cells with prolonged use. In the present study, the somatic mutation and recombination test (SMART) was employed to determine the mutagenic effects of Pantoprazole on Drosophila melanogaster. Repeated treatments with Pantoprazole were performed on 72-hour larvae of the standard (ST) and high bioactivation (HB) crosses at concentrations of 2.5, 5.0, and 10.0 μM. In addition, doxorubicin (DXR) was administered at 0.4 mM, as a positive control. When administered to ST...
